ABSTRACT

The transcription factor ZNF24 (also known as ZNF191 or KOX17) is a member of the Krüppel-like zinc finger transcription factor family, with a leucine-rich (Leu) SCAN domain (also known as LeR domain) at the N-terminus and four consecutive typical Kruppel-like zinc finger modities at the C-terminus. ZNF24 is a multifunctional transcription factor involved in the regulation of kinase transcriptional activity, vascular proliferation and development, especially in tumorigenesis and tumor progression. ZNF24 plays an important and complex dual-directional regulation role (promoting and inhibiting) in tumor development, invasion and metastasis by regulating the transcriptional expression of different target genes (such as VEGF, Wnt8B, Twist1, β-catenin and DGL1, etc.) and the competitive binding with protein factors (such as β-catenin). Therefore, elucidating the mechanism of ZNF24 in tumors would provide clues and ideas for the treatment of tumors. The researches status of ZNF24 in tumors were summarized in this paper.

ABSTRACT

Podocyte infolding glomerulopathy (PIG) is a pathologic type of podocyte glomerulopathy reported recently. The characteristic is that the ultrastructure related to podocytes, such as microspheres and microtubules, are folded into the glomerular basement membrane (GBM) under electron microscope. At present, there are few reports about this disease at home and abroad, and most of them are concentrated in Japan. The clinical characteristics and pathogenesis of PIG are still unclear. In this paper, we report a case of clinical manifestations of nephrotic syndrome, renal biopsy indicated PIG, after the treatment of glucocorticoid, hydroxychloroquine and tacrolimus, the patient's clinical symptoms were relieved and urinary protein decreased.

ABSTRACT

Objective:To investigate the antimicrobial properties of copper-loaded coatings on the surface of ureteral stents and their biocompatibility in order to determine the most suitable level of copper loading.Methods:Copper-loaded PDA coatings with different copper contents were constructed on the surface of polyurethane (PU) stents using polydopamine (PDA) and dimethylaminomethylborane (DMAB). The antibacterial property of the coating against Escherichia coli and Staphylococcus aureus was investigated by the plate counting method. The bacterial adhesion on the coating surface was studied by scanning electron microscopy. Using the live/dead evaluation, microbes were stained and observed by a fluorescence microscope. The biocompatibility of the copper-loaded coatings was detected by a cell proliferation assay incubated with L929 cells. Results:The antibacterial rates of the copper-loaded samples exceeded 90% after incubation with E. coli and S. aureus for 24 h, respectively, and the antibacterial performance increased with the increase of copper content in the coating. The amount of bacteria adhered to the surface of the copper-loaded samples was significantly lower, and most of them were dead bacteria. When the copper content in the coating preparation solution used was 0.25~1 g/L, the cell proliferation rate on the surface of the copper-loaded coating was higher than 80% and the material was not cytotoxic. Conclusions:A copper-loaded PDA coating with excellent antibacterial properties and good biocompatibility can be prepared with a copper content of 1 g/L in the coating preparation solution, forming a potential solution for the preparation of ureteral stent coatings.

ABSTRACT

Objective:To investigate the prevalence of chronic kidney disease(CKD)in community of Suzhou and explore the comorbid health conditions. Methods:The population over 60 years old undergoing health physical examination in community of Suzhou were selected as screening subjects for CKD.Examination data were collected, and other health data were obtained through residents' health records and chronic disease management systems.A total of 7 387 participants were enrolled. Results:In the population of 7 387 individuals, detected cases with CKD were 2 771, with detection rate of 37.5%, awareness rate of 13.9%.The 2 521 patients(91.0%=2 521/2 771 of total CKD patients)with CKD 1-2 stage, and 2222 patients(80.2%=2 222/2 771)with low and medium risk were detected out.The detection rates of CKD were significantly higher in group with hypertension, diabetes, hyperuricemia, hyperlipidemia, obesity and stroke than in the group without those diseases(all P<0.05). With the increase of the number of comorbidity in patients with CKD, the detection rate of CKD was increased( χ2=74.658, P<0.001). In a total of 2771 CKD patients, 33.6% had 1 comorbid condition, 23.4% had two comorbid conditions, and 19.8% had 3 or more comorbid conditions. In the declined-eGFR group, patients with three or more comorbid conditions accounted for 34.0%, which was significantly higher than that in the normal-eGFR group(18.4%)( χ2=35.042, P<0.001). Multivariate Logistic regression analysis revealed that women, advanced age, hypertension and having 3 or more comorbid conditions were independent influencing factors for the CKD detection rate.People with three or more comorbid conditions had a 1.58-fold increased risk for developing CKD as compared with those without comorbid condition(95% CI: 1.30-1.91, P<0.001). Conclusions:The prevalence of CKD in Suzhou community elderly population is high, but the awareness rate is low.A large number of early and low-and moderate-risk of CKD patients have been detected out.Comorbid conditions are prevalent in older patients with CKD.With the decrease of renal function, the number of comorbidity in CKD patients significantly increased.Early screening for CKD in the elderly population is very necessary, especially in those with hypertension and 3 or more comorbid conditions.

ABSTRACT

In this article, based on z-curve theory and position weight matrix (PWM), a model for nucleosome sequences was constructed. Nucleosome sequence dataset was transformed into three-dimensional coordinates, PWM of the nucleosome sequences was calculated and the similarity score was obtained. After integrating them, a nucleosome feature model based on the comprehensive DNA sequences was obtained and named CSeqFM. We calculated the Euclidean distance between nucleosome sequence candidates or linker sequences and CSeqFM model as the feature dataset, and put the feature datasets into the support vector machine (SVM) for training and testing by ten-fold cross-validation. The results showed that the sensitivity, specificity, accuracy and Matthews correlation coefficient (MCC) of identifying nucleosome positioning for were 97.1%, 96.9%, 94.2% and 0.89, respectively, and the area under the receiver operating characteristic curve (AUC) was 0.980 1. Compared with another z-curve method, it was found that our method had better identifying effect and each evaluation performance showed better superiority. CSeqFM method was applied to identify nucleosome positioning for other three species, including , and . The results showed that AUCs of the three species were all higher than 0.90, and CSeqFM method also showed better stability and effectiveness compared with iNuc-STNC and iNuc-PseKNC methods, which is further demonstrated that CSeqFM method has strong reliability and good identification performance.

ABSTRACT

Objective To investigate the curative effect of lateral position ventilation combined with vibration sputum drainage on the patients with acute respiratory distress syndrome (ARDS). Methods A prospective randomized controlled trial was conducted. The patients with ARDS undergoing mechanical ventilation (MV) admitted to intensive care unit (ICU) of Linyi Central Hospital from January 2013 to June 2017 were enrolled, and they were divided into simple ventilation group and combined treatment group according to random number table. The patients in both groups received etiological treatment, protective ventilation strategy, sensitive antibiotics for anti-infection, and calefacient and humidifying treatment. The patients in the simple ventilation group received bilateral discontinuous alternative lateral position ventilation [pressure controlled ventilation (PCV), tidal volume (VT) ≤ 6 mL/kg, the inhaled oxygen concentration (FiO2) and positive end expiratory pressure (PEEP) was adjusted to maintain the airway plateau pressure (Pplat) ≤ 30 cmH2O (1 cmH2O = 0.098 kPa)], and those in the combined treatment group received lateral position ventilation combined with vibration sputum drainage, twice a day, 15 minutes each time. The parameters of respiratory function and inflammation as well as excretion of sputum before and after treatment were compared between the two groups, and the complication and prognosis were recorded. Results A total of 200 patients with ARDS were included, 4 patients were excluded because of severe pneumothorax, massive hemorrhage of the digestive tract, or elevated intracranial pressure, so 196 patients were finally enrolled in the analysis. There were 98 patients in the simple ventilation group and the combined treatment group, respectively. There were no significant differences in parameters of respiratory function and inflammation as well as excretion of sputum before treatment between the two groups. After treatment, the above parameters in both groups were improved, and the effect of combined treatment was more significant. Compared with the simple ventilation group, white blood cell count (WBC), procalcitonin (PCT), C-reactive protein (CRP) at 24 hours of treatment in the combined treatment group were significantly decreased [WBC (×109/L):9.1±1.6 vs. 11.8±3.6, PCT (μg/L): 14.5±2.4 vs. 22.7±3.2, CRP (mg/L): 32.2±6.3 vs. 67.2±7.2, all P < 0.01], the quantity of sputum excretion was significantly increased (mL: 49.3±12.5 vs. 36.8±11.8, P < 0.01); 72 hours after treatment, the oxygenation index (PaO2/FiO2) in the combined treatment group was significantly increased [mmHg (1 mmHg = 0.133 kPa): 278±28 vs. 238±39, P < 0.01], and PEEP, FiO2, airway resistance (Raw) were significantly lowered [PEEP (cmH2O): 5±2 vs. 7±3, FiO2: 0.40±0.11 vs. 0.49±0.12, Raw (cmH2O): 12.8±0.7 vs. 16.2±0.8, all P < 0.01]. There was no significant difference in static lung compliance (Cst) or Pplat between the two groups. Compared with the simple ventilation group, the duration of MV (days: 5.5±3.0 vs. 8.6±2.1), the length of ICU stay (days: 7.5±5.4 vs. 11.3±4.6) and the extraction time of artificial airway (days: 6.6±2.8 vs. 9.4±3.3) in the combined treatment group were significantly shortened (all P < 0.01). However, there was no significant difference in the 28-day mortality rate [4.1% (4/98) vs. 3.1% (3/98)], the rate of tracheotomy [2.0% (2/98) vs. 3.1% (3/98)] or the incidence ofⅠ-Ⅱ skin pressure sore [1.0% (1/98) vs. 2.0% (2/98)] between simple ventilation group and the combined treatment group (all P > 0.05). No artificial airway disappearance, pneumothorax, grade Ⅲ or Ⅳ pressure sore was found in the two groups. Conclusions Compared with lateral position ventilation alone, the treatment of combined with vibration sputum drainage is more effective for improving the respiratory function of patients with ARDS, reducing infection action, shortening the duration of MV and the length of ICU stay, and improve the recovery, promote the rehabitation of patients.

ABSTRACT

OBJECTIVE@#To investigate the phenotype and genetic characters of a Chinese family with an autosomal-dominant inherited high-frequency sensorineural hearing loss.@*METHOD@#A Chinese pedigree associated with an autosomal-dominant inherited high-frequency sensorineural hearing loss was investigated. After obtaining informed consent from all study participants medical and audiological examination to rule out any syndromic hearing impairment. Application of microsatellite markers on DFNA 21 loci preliminary screening of 23 genes, data were analyzed by linkage analysis.@*RESULT@#Proband of the family had been diagnosed with high-frequency sensorineural hearing loss. A Chinese family SX-H043 with non-syndromic hearing loss were ascertained. This Chinese family with late onset hearing impairment spanned four generations and comprised 43 members. The mode of inheritance of the families should be autosomal dominant according to the pedigree. Hearing impairment of affected members in family SX-H043 occurred 25 to 50 years descending audiograms. Whole frequencies became involved with increasing age.@*CONCLUSION@#A Chinese family with late-onset high-frequency sensorineural hearing loss were clinically studied. Positive sites were not found in the known deafness loci screening. The information should facilitate future gene scan and linkage analyses for novel relative genes contributing to high-frequency sensorineural hearing loss.

Objective To investigate the independent risk factors of cerebral white matter lesions (WML) of different degrees in the elderly aged 80 years and over, and provide the evidences for forecasting the prognosis of WML.Methods Brain magnetic resonance images (MRI) findings in 151 people aged 74 to 93 years were collected and analyzed.According to the severity of WML in brain MRI using the Fazekas Scale, the persons were divided into non-WML (control) group, mildWML (grade 1 WML) group and moderate-to-severe WML (grade 2 WML) group. The cognitive score, vascular risk factors, cerebral hemodynamic and arteriosclerotic index, and radiological features were compared among the three groups.Subsequent one-way ANOVA and multivariate logistic analysis were performed to determine the statistically significant factors and the independent risk factors among groups. Results The statistically significant factors with one-way ANOVA analysis among the three groups were cognitive performance (F = 48.595,P = 0.000),hypertension (x2 =7.052,P=0.029),cigarette history (x2 = 19.476,P= 0.000),cholesterol (TC) (F= 3.086,P=0.049),Crouse score (F=3.968,P=0.021) and multiple cerebral atrophy indexes. When compared with control group, cigarette history (OR 2.031,95%CI 1.244-1.317),lacunar infarction (LI)numbers (OR 2.031,95%CI 1.316-4.015) and cholesterol (OR 1.610,95%CI 0.972-2.668) were independent risk factors in grade 1 WML group (all P<0.05).The independent risk factors between grade 1 and 2 WML group were cognitive performance (OR 0.276,95%CI 0.143-0.532),cigarette history (OR 2.262,95% CI 1.260-4.059),and sylvian fissure ratio (SFR) (OR 1.954,95% CI 1.013-3.768) (all P<0.05).The independent risk factors between the grade 2 WML group and control group were cognitive performance (OR 0.091,95%CI 0.030-0.273),bicoudate ratio (BCR)(OR 2.511,95%CI 1.147-5.499),Crouse score (OR 2.304,95%CI1.127-4.712)and LI numbers (OR 2.200,95%CI 1.028-4.707) (all P<0.05).Conclusions Mild WML patients have no significant abnormalities in cognition, brain atrophy and cerebral atherosclerosis.Moderate to severe WML patients manifest remarkable cognitive disorder, cerebral atherosclerosis and brain atrophy.Compared with the controls, cognitive performance,BCR,Crouse score,LI numbers were the independent risk factors for moderate-severe WML patients.

ABSTRACT

Different geographic seed sources (80) of Jatropha curcas L. were collected in South China and planted in a germplasm resource garden to study their biological and agricultural properties. The average ground diameter, tree height and crown size of two-year old plants of the 80 sources was 7.6 cm, 167 cm and 114 cm, respectively, the average 1000-seed weight was 0.676 (0.477-0.876) kg. The trees grew further to the average size of 12.6 cm diameter, 2.69 m height and 2.1 m crown at the 4th year. Among the 80 sources, six sources had higher oil yield (seed oil content of 40%-42%) and better behaving in expression of phenotype were selected for a small-scale trial of forestation to determine oil yield. Among them a provenance with outstand in expression of phenotype yielded 964.3, 2000.6 and 2858.7 kg/ha was achieved for two- three- and four-year old trees, respectively. Additionally, a new Jatropha mutant was found in the wild and hybridization experiments showed that its oil content increased by 6%.

ABSTRACT

Objective To investigate the phenotype and genetic characteristics of a Chinese family with an autosomal-dominant inherited sensorineural hearing loss. Methods A Chinese pedigree associated with an autosomal-dominant inherited low-frequency sensorineural hearing loss (LFSNHL) was investigated. After obtaining informed consent from all study participants medical and audiological examination were used to rule out any syndromic hearing impairment.Five patients were tested with DPOAE and ABR,while two patients were tested with vestibular function and computed tomography scan of the temporal bone to exclude auditory neuropathy and other possible aural disorders.Twenty-one loci and twenty-three genes of DFNA screening had been done by using microsatellite markers and linkage analysis. Results Proband of the family had been diagnosed with low-frequency sensorineural hearing loss.A Chinese family BJ-L046 with non-syndromic autosomal dominant hearing loss was ascertained.Hearing impairment in the affected members in family BJ-L046 occured from 10 to 20 years of age and mainly affected the low frequencies, causing an upsloping audiogram.Higher frequencies were getting involved with increasing age, thus causing a flat-type audiogram. No positive result was found in twenty-one loci and twenty-three genes of DFNA screening. Conclusion A Chinese family with late-onset low-frequency sensorineural hearing loss was clinically studied. No positive result was found by linkage analysis, and twenty-one loci and twenty-three genes of DFNA were preliminary excluded.

ABSTRACT

Objective: To observe the therapeutic effect of acupuncture therapy for treatment of chronic atrophic gastritis. Methods: According to the theory of TCM, 36 cases of chronic atrophic gastritis were differentiated and treated by acupuncture. Gastroscopy and electrogastrograph (EGG) of the body surface, gastric acid and pepsin, blood gastrin, blood prostaglandin E (PGE), gastric mucosal cells and G cells are recorded. Results: Acupuncture therapy had a better effect in improving clinical symptoms, the markedly effective rate was 97.2% and the disappearing rate of clinical manifestations 97.02%;after acupuncture treatment, blood PGE concentration increased; gastric acid secretion increased; gastric dynamics was enhanced and the amplitude of EGG rose; acupuncture could significantly facilitate secretion of gastrin. Conclusion: Acupuncture is a convenient treatment method for atrophic gastritis with quick effect and without side effects.
